Abstract
A simple, rapid and reliable high performance liquid chromatographic method for simultaneous determination of six quinolones (cinoxacin, nalidixic acid, pipemidic acid, ofloxacin, pefloxacin and ciprofloxacin) has been developed. Isocratic separation was achieved using a Nacalai Cosmosil 5C18-MS column ( 25cm x 4.6 mm I.D.) with a mobile phase consisting of 3mM sodium lauryl sulfate in methanol : 40mM phosphate buffer : acetonitrile (25:55:20) pH=3.50. An ultraviolet absorption photometer was used as the detector at the wavelength of 257nm. The HPLC separation was completed in 40 min, giving an excellent resolution and peak shape. Piroxicam was used as the internal standard for the assay. The precision, accuracy and reproducibility were satisfactory. The system can be used for assay of six quinolones in dosage forms as well as for identification of these six quinolones in unknown samples.
